DNA Sequencing at BTI

DNA sequencing is now available on a first-come/first-served basis using a state-of-the-art ABI Prism 3100 Genetic Analyzer/ DNA Sequencer. This machine is capable of sequencing 16 samples simultaneously with autoloading from 96- or 384- well plates. All you need to provide is a completed BTI DNA Sequencing form (PDF | MS Word) and a sample prepared using the required DNA Sequencing Protocols. The form and the protocol sheet can either be downloaded, found in the BTI Library, or picked up at the sample drop-off station.

The cost for each sample run will be $2.75. Reagent costs are about $2.50 per sample. The samples are prepared similarly to the methods used for the ABI 310 or 377. There are a couple of alterations, however, so be sure to follow the DNA Sequencing Protocol for best results.

Due to the high cost of the ABI Big Dye Kit, 100 µl aliquots are available in the stockroom (enough for ten or more reactions, depending on the amount of template used). DO NOT order these kits yourself!!! The stockroom has bought this reagent in bulk, and therefore is able to offer end users a price that reflects a SIGNIFICANT discount!!! The Qiagen miniprep kit must be ordered by each lab individually.

Please be sure that your samples are clearly marked, and that each is uniquely labeled (i.e. R43-S, NOT 1,2,3,4). See DNA Samples (or see Steve) for information on where to place your prepared samples. When you drop off your samples, leave your completed DNA Sequencing Form in the folder marked "Completed DNA Sequencing Forms BTI" located on the refrigerator. On the form, be sure that you include your name, sample ID number, and project number that the run(s) should be billed to, or your samples will not be run (I can usually figure out the rest). Samples will be processed as demand dictates. You can access your analyzed data from your computer as soon as your samples have been run; I will send an e-mail describing the sample folder where your data can be found. See Steve or Diana regarding pathways and passwords to sample folders. Data return is taking about 1-3 days at this point.
